Recently, many cross contaminations are rising in the human cell lines. Therefore, the certification of the cell line used in the laboratory is expected to be essential for the trustful result of the research. The case is increasing that the certification of the cell line is recommended or required in the submission of paper year by year, i.e., Nature requires STR data when the authors establish ES cells. In addition, clearly requiring the cell certification for publishing paper are the magazines American Association for Cancer Resaerch (AACR) publishes including Cancer Rearch, the magazines Springer publishes including In Vitro Cellular & Developmental Biology、Cell Biochemistry and Biophysics, and the magazines Wiley publishes including International Journal of Cancer.
As the method to certificate the cell line, a cell bank in US, ATCC suggests microscoping morphology, analysis of growth curve, Isozyme analysis (estimating species), STR(Short Tandem Repeat). In these methods, STR method is the most used from the point of view of recognizability and objectivity. STR method was, originally, developed as a method to recognize the indivisual in forensic research. That's why STR is very effective for certification of the cell line.
STR Analysis is spotlighted as the easiest method to certificate the cell lines. In GenePrint® System (Promega), specific locus in human genome is analyzed. amplified and detected by 3 kinds of fluorescene are 10 locus (9 STR locus + 1 amelogenin gene for gender detection: D21S11,TH01,TPOX,vWA,Amelogenin,CSF1PO,D16S539,D7S820,D13S317, and D5S818). The detection is improved by addition of D21S11 locus to PowerPlex1.2® System.
PowerPlex16® System (Promega) is often used to check the genotype of iPS / ES cells.
Amplified at the same time and detected by 3 fluorescenes are 16 locus: 6 fluorescene-labelled locus (D3S1358,TH01,D21S11,D18S51,Penta E and Amelogenin), 4 Tetramethylrhodamine-labelled locus (VWA,D8S1179,TPOX,FGA), 6 JOE-labelled locus (5S818,D13S317,D7S820,D16S539,CSF1PO,Penta D).
